Differentiation of pathogenic and non-pathogenic isolates of Philippine Entamoeba spp. by the Polymerase Chain Reaction.

Fecal material was inoculated into different growth media. Some of the isolates adapted well in Robinson's medium while the others grew better in Inoki's medium. Of the 210 stool specimens collected, 15 isolates were successfully cultured. After mass cultivation in axenic cultures, 10 exponent 6 cells were harvested and genomic DNA extracted by alkaline lysis method. Primers known to be specific for pathogenic and non-pathogenic strains were used to characterize the amoeba isolates. Amplified DNA segments were digested with restriction enzymes that could distinguish between pathogenic and non-pathogenic forms. Thus far, 4 of the local isolates have been confirmed as pathogenic E. histolytica and 2 are non-pathogenic E. dispar. (Author)
